Having built the long block and primed the oil pump. I see, from timing the cams, that the engine goes through quite a few revolutions. Won't this displace all the assembly lube inside the oil pump? Does this mean I need to reprime my oil pump?

When you're tuning the engine over by hand it's turning too slowly to really do much to the lube that you've packed the pump with so you don't need to re-prime the pump.
