This seems to make sense to me, but want to check I'm not overthinking it!

I want to have a Relay (100amp) that activates power to the fuse box when the ignition is switched on (Key Switch).

So I'm thinking of running, cable from the starter to the fuse box, with the relay in the middle, this is activated by the switch. Thus meaning for power to work to the fuse box the key has to be in and turned on.

Is this logical?

It's hard to give advice not knowing your exact implementation; EG, is this a road car/race car and is it the main fuse board or a sub fuse board? If it's a subboard, a main board, and a race car, maybe a safety kill switch would be a better option. If it's a road car, maintain any permanent lives that might be required for rego or MOT, Hazards etc.

I just did two sub-fuse boards in this way: one is running a switched live for audio and the other for the air ride. In most cases, the battery and the starter are electrically connected, so either would work. Just make sure you aren't circumventing any fusing or that you are adding a main fuse for that board.
