For high-vacuum cells that are very difficult to reach on the road (for example, cells that only occur when going downhill with very little throttle opening), is it acceptable to tune the fuel in those cells with the vehicle stationary and no load on the engine? Or will the fuel requirements be significantly different compared to when the vehicle is actually driving?

It can get you close enough to validate and fudge but don't tear your hair out trying to stick to your zero load/free rev numbers if datalogs are telling you differently - proper VE numbers will probably be somewhere in the middle.
